It sounds like we have a door lock actuator sticking. You will need to pull the rear door panel off and verify the noise is coming from the door lock actuator (it's right near the latch). Disconnect the wiring harness from the actuator and this will confirm your noise. It sound like a simple door lock actuator replacement.

We’ve identified that it is coming from inside the door. Do we still need to pull the panel apart ?

I will be glad to call you but I don't think it's necessary and save your money to purchase the new actuator. If the noise stops when you press the lock button it's definitely the door lock actuator. The door panel will need to come off so it can either be replaced or disconnected to stop the noise. The noise should be due to the door lock actuator "actuating". This really should be the only moving piece inside the door besides the window motor, which should not be affected by the door lock. But to confirm we need to pull the door panel off either way.
